I was thinking of unpublishing my eBook temporarily, then putting it back up. Will this effect those who have already bought it?

It can get you in trouble with Amazon as it would be putting your book on the newly published list twice.
If you want to make some changes to the book, just upload a corrected version OVER what is there now.
To answer your question in your header. ... YES, assuming you mean possible new buyers.

Actually people who have bought your book still have it, even if you unpublish. (This is why you can't totally delete an ebook if it has had sales--it has to still exist for the benefit of previous buyers.)

But as Donna says, there's no reason to unpublish if you just want to make some updates.

Except in very unusual circumstances (a bandit copy of 1984 or maybe it was Animal Farm comes to mind), an ebook once purchased is always available to the purchaser.

(And in that case where the book was removed from purchasers' bookshelves, they were refunded the purchase price.)

(Don't trust KDP to publish a print edition. Don't trust CreateSpace to publish an ebook. Each does one thing well and the other thing poorly.)

Good luck! -- NJ
