This is my first book kindle book I have published. I would like to get some feedback and ideas how to increase the sale, the sales are almost 0. Only my friends know about this book at the moment.

It is online since 6 days.

1) Does using the 5 day promo by amazon help to boost the sale? I heard a lot of people get into top 100 but only in the 'free kindle book' section and not in the paid one

Does this mean my paid sales rank will still be low desptie of the free promo?

I haven't looked at your book--but prepare yourself, because those that give feedback try to tell you honestly where the problems are, rather than praising you and telling you how everything is wonderful.

Don't do the give-the-book-away-for-5-days thing if it's your only book. All that happens is that anyone who might have been willing to spend money on it now owns it for free. And while you may get an upward blip in the "free" rankings, your rank among paid books will sink from sight. The two sets of ranks have no effect on each other, and if you have 5 days with only freebies, then anyone who sold a book for money during those 5 days will pull ahead of you in rank.

Welcome to KDP! It's a lot of fun, but selling your book is a lot harder than writing it.

In my experience, people looking for freebies don't buy books. They just wait for the next freebie.

And with only one book, how can you benefit by giving the book away? Probably the only time a freebie pays off is for the writer of a series of books, hoping people will get hooked on the series.

(Don't trust KDP to publish a print edition. Don't trust CreateSpace to publish an ebook. Each does one thing well and the other thing poorly.)

You're up against a lot of competition, thousands and thousands of books, all with captivating covers, intriguing blurbs, scintillating narration and dialogue that is realistic and properly formatted. I think your book (actually, at only 80 pages, I'm reluctant to use that term) needs more work before its truly competitive with all those other books. As to giving away your only book, you only succeed in losing customers.

You have an interesting premise but your book is too short and there are obvious grammatical errors that suggest English is not your native language. Both issues can be fixed.

Do not give your book away. Take it down and write about 50 thousand to 60 thousand words. But no filler/padding. You are aiming to write like Michael Creighton.

Write a stellar book and get it edited. Then publish it.
