Once you've finished publishing and your book appears "live" on your bookshelf (can take up to three days) you click on the select box by the title you want to edit, and then in the drop-down options you can edit anything about the book at all, including uploading an entire new file if you've revised it. If you've made significant revisions consider changing the edition number on the book details. Then just republish. DON'T publish as a new book if you're just making amendments, just edit the original.
If you're only changing the description (blurb) you can do this through author central http://authorcentral.amazon.com and the changes will go live in about 15 mins. To change the book file, cover image or price you need to go through the bookshelf and it will take 1-3 days to roll out completely. It will overwrite the old file and the book will remain available for purchase throughout.
